APOE4 is the most significant genetic risk factor for late-onset Alzheimer's Disease. The N-terminal domain (1–167) carries LDL-receptor binding region while C-terminal domain has lipid binding region. Amino acids at 112, 158 give isoforms (E2, E3, E4).
